Background:
Nosema apis and Nosema ceranae are microsporidian parasites recently reclassified as a fungus. Both species cause nosema disease in honeybees (Apis mellifera). The dormant stages are long-lived spores. The most notable symptom is dysentery. Nosema ceranae can be detected in all four seasons, while Nosema apis occurs mostly in the milder seasons of autumn and spring. There is a higher mortality of bees when they are infected by Nosema ceranae than when infected with Nosema apis. Double infection is also possible.
Description:
MycoReal® Kit Nosema apis & ceranae uses multiplex real-time PCR for the qualitative detection and differentiation of the 18S rRNA gene of Nosema apis as well as of the 18S rRNA gene of Nosema ceranae. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms:
MycoReal® Nosema apis & Nosema ceranae was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), LightCycler® 480 II (Roche Diagnostics) and Mx3005P® (Agilent). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in FAM and Cy5 channel (eg. QuantStudio™ 5/6/7 Real-Time PCR System (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), cobas z 480 Analyzer (Roche)).
Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

It detects the 18S rRNA gene of Nosema ceranae (FAM channel) and Nosema apis (VIC channel) using real-time PCR.
Infection with Nosema ceranae or N. apis leads to nosema disease in honey bees, causing colony weakening and reduced survival.
The LoD95% is 1.06 copies/reaction for N. ceranae and 1.46 copies/reaction for N. apis.
A DNA internal positive control (IPC) is included to monitor extraction and PCR performance, detected in the Cy5 channel.
An assay contains detection reagents only, while a kit includes additional components like mastermix, buffers, and workflow tools.
