Background:  The family Coronaviridae includes pathogenic porcine coronaviruses such as the transmissible gastroenteritis virus (TGEV), the porcine respiratory coronavirus (PRCV), the porcine epidemic diarrhea virus (PEDV) and the porcine hemmaglutinating encephalomyelitis virus (PHEV). TGEV causes transmissible gastroenteritis (diarrhea and vomiting) in pigs, mortality is highest in neonates. PRCV is a mutant of TGEV and does not appear to be an important primary pathogen.

Description:  ViroReal® Kit TGEVs is a real-time PCR for the qualitative detection of a part of the S region of the Transmissible Gastroenteritis Virus (TGEV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The test does not detect PRCV that has a large in-frame deletion in the 5’ end of the S gene. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit TGEV was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), and cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The test amplifies the S gene region of TGEV RNA using a probe detected in the FAM channel.
The RNA IPC checks for RNA extraction success and RT-PCR inhibition, ensuring the validity of negative results.
The kit is validated for ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, qTOWER3G, MIC, and cobas z 480.
Primers and probes were validated against public sequences and tested to ensure specific detection of TGEV without cross-reactivity to PEDV, PRRSV, PCV2, or PHEV.
If the IPC is not added during extraction, it can be added to the PCR master mix to monitor only the RT-PCR reaction.
