Background:  Streptococcus uberis is a Gram-positive bacterium known worldwide as an environmental pathogen responsible for a high proportion of cases of clinically acute as well as subclinical bovine mastitis.

Description:  BactoReal® Kit Streptococcus uberis is a real-time PCR for the qualitative detection of ITS region of S. uberis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Streptococcus uberis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit amplifies the ITS region of Streptococcus uberis DNA, with detection measured in the FAM channel of real-time PCR instruments.
S. uberis is a Gram-positive bacterium and one of the main environmental pathogens causing both acute and subclinical bovine mastitis, leading to economic losses in dairy farming.
The kit is validated for DNA extracted from milk and other materials relevant for bovine mastitis testing.
An internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, monitors extraction efficiency and detects potential PCR inhibition.
The analytical sensitivity is 1 target copy per PCR reaction, with a limit of detection (LoD95%) of 9 copies. No cross-reactivity was observed with other tested bacterial species.
