Background:  Staphylococcus aureus is a gram-positive bacterium. In healthy animals, S. aureus colonization of the skin, intestinal tract, or nasopharynx is generally asymptomatic. However, depending on the strain, S. aureus is capable of secreting several exotoxins. Many of these toxins are associated with specific diseases. By producing a toxin, S. aureus is also a common cause of foodborne illness. In cattle, S. aureus causes one of the most common types of chronic mastitis. In humans, S. aureus is a frequent commensal of the skin, but can cause a range of illnesses from minor skin infections to life-threatening diseases such as pneumonia, meningitis, osteomyelitis, endocarditis, toxic shock syndrome and sepsis.

Description:  BactoReal® Kit Staphylococcus aureus is a real-time PCR for the qualitative detection of the thermonuclease (nuc) gene of S. aureus. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Staphylococcus aureus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of Staphylococcus aureus DNA by targeting the thermonuclease (nuc) gene using real-time PCR.
S. aureus is a commensal bacterium but can cause diseases ranging from skin infections to pneumonia, meningitis, osteomyelitis, toxic shock syndrome, and sepsis. In cattle, it is a common cause of chronic mastitis.
Validated samples include DNA extracts from swabs, biopsies, and milk.
An internal DNA positive control (IPC) is added during extraction and detected in the Cy5 channel, verifying extraction efficiency and excluding PCR inhibition.
The analytical sensitivity is 5 copies per reaction with a LoD95% of 30 copies. No cross-reactions were observed with tested non-target bacteria.
