Background:  Schmallenberg virus (SBV) is a novel Orthobunyavirus that has been associated with disease in ruminants (cattle, sheep and goats) in Europe. It causes transient clinical symptoms in adult cattle and congenital malformation in newborn ruminants. Schmallenberg virus is part of the Simbu serogroup of the Bunyaviridae family, genus Orthobunyavirus. The virus is transmitted via insect vectors, mainly midges.

Description:  ViroReal® Kit Schmallenberg Virus is a real-time PCR for the qualitative detection of the RNA of the nucleocapsid protein gene of Schmallenberg virus (SBV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Schmallenberg Virus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas z 480 Analyzer (Roche), and QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The test targets the nucleocapsid (N) gene of SBV RNA, detected via fluorescence in the FAM channel.
The test may detect other Simbu serogroup viruses like Shamonda, Sathuperi, and Douglas virus, due to sequence similarities.
The IPC-Target RNA is added to the lysis buffer before RNA extraction to validate both the extraction and amplification steps.
Validated systems include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Use separate workspaces for RNA extraction and amplification, use RNase-free materials, and pipette the positive control last.
