Background:  Salmonella is a genus of Gram-negative bacteria of the Enterobacteriaceae family. This genus consists of only two species, Salmonella enterica and Salmonella bongori. Salmonella bongori mostly infects cold-blooded animals. Salmonella enterica has 6 subspecies and more than 2500 serovars. Subspecies are S. enterica subsp. arizonae, S. enterica subsp. diarizonae, S. enterica subsp. enterica, S. enterica subsp. houtenae, S. enterica subsp. indica and S. enterica subsp. salamae. Salmonella infections in warm-blooded animals are generally caused by strains from S. enterica subspecies enterica. Nontyphoidal serovars are more common, and usually cause self-limiting gastrointestinal disease. They can infect a range of animals, and are zoonotic. Although many other Salmonella serovars may cause enteric disease, the more common ones in cattle are Salmonella Typhimurium, Salmonella Dublin and Salmonella Newport. In sheep and goat the most common are Salmonella Typhimurium, Salmonella Dublin, Salmonella Abortusovis, Salmonella Anatum and Salmonella Montevideo. In pigs the most common are Salmonella Typhimurium and Salmonella Choleraesuis, and in poultry Salmonella Enteritidis, Salmonella Typhimurium, Salmonella Gallinarum and Salmonella Pullorum.

Description:  BactoReal® Kit Salmonella enterica is a real-time PCR for the qualitative detection of the invasion protein A (invA) gene of Salmonella enterica. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Salmonella enterica was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects DNA of Salmonella enterica by targeting the invA gene with real-time PCR and fluorescence measurement in the FAM channel.
S. enterica includes more than 2500 serovars that cause enteric disease in humans and animals. Common serovars in livestock include S. Typhimurium, S. Dublin, S. Choleraesuis, and in poultry S. Enteritidis.
The kit is validated for DNA extracts from swabs and faecal samples of animals.
An internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, ensures proper DNA extraction and identifies PCR inhibition.
The analytical sensitivity is 7 DNA copies per PCR, with a LoD95% of 19 copies per reaction. Validation confirmed specificity for S. enterica; S. bongori is detected with reduced sensitivity.
