Background:  The sacbrood virus (SBV) is a member of the family Iflaviridae. The genome is a single-stranded, positive-sense RNA molecule. Sacbrood affects the brood of the honeybee, resulting in larval death. Larvae are infected by consuming brood food contaminated with SBV. Larvae with sacbrood fail to pupate and the skin of the larvae then gradually becomes a fluid filled sac. The SBV may remain viable in dead larvae, honey or pollen for up to several weeks. SBV may also affect the adult bee, but without symptoms. Sacbrood occurs most frequently in spring.

Description:  ViroReal® Kit Sacbrood Virus is a real-time PCR for the qualitative detection of the RNA of the polyprotein gene of Sacbrood Virus. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Sacbrood Virus was validated with the QuantStudio™7 Real-Time PCR System (Thermo Fisher Scientific) and the QuantStudio™5 Real-Time PCR System (Thermo Fisher Scientific). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in FAM and Cy5 channel (eg. QuantStudio™5/6/7 Real-Time PCR System (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio moleclar systems), cobas z 480 Analyzer (Roche)).

The kit enables qualitative detection of RNA from Sacbrood Virus (SBV) by targeting the polyprotein gene using reverse transcription real-time PCR.
SBV is a viral pathogen of honeybee larvae. Infected larvae fail to pupate and develop a fluid-filled sac, which leads to larval death and affects colony health.
An artificial RNA internal positive control (IPC) is added during extraction or PCR to monitor RNA extraction efficiency and detect RT-PCR inhibition, detected in the Cy5 channel.
The kit is validated for RNA extracted from honeybee larvae or adult bees.
The LoD95 is 14.15 target copies per reaction based on validation with synthetic RNA dilutions.
