Background:  Pseudorabiesvirus (PRV) is a swine DNA herpesvirus of the <i>Alphaherpesvirinae </i>subfamily. It is also known as suid herpesvirus 1 (SHV-1), or by its original name, Aujeszky''s disease virus. The pig is the principal reservoir host, but the virus may also infect other mammals like cattle, little ruminants, carnivores, and rodents. It is not pathogenic for humans. In swine, Aujeszky’s disease shows itself under three forms: a nervous form, a respiratory form, or a genital form. In latently infected pigs, pseudorabiesvirus persists primarily in the trigeminal or sacral ganglia and the tonsils. In other mammals, it causes infection of the central nervous system with neurologic behavioral changes leading very quickly to death.

Description:  ViroReal® Kit Pseudorabiesvirus is a real-time PCR for the qualitative detection of the DNA of the glycoprotein B gene of Pseudorabiesvirus (SHV-1). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Pseudorabiesvirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas z 480 Analyzer (Roche), and QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit amplifies the glycoprotein B gene of PRV DNA, detected in the FAM channel.
Clinical signs include fever, neurological disorders, respiratory disease, and reproductive problems. In other mammals, PRV infection leads to severe CNS involvement and rapid death.
The IPC ensures proper DNA extraction and detects RT-PCR inhibition, thereby reducing the risk of false-negative results.
