Background:  Pasteurella multocida is a Gram-negative bacterium and part of the flora in the respiratory tract of pigs. The bacterium induces hemorrhagic septicemia or, usually as a secondary pathogen invading lungs injured by other bacteria or viruses, pneumonia. A subset of P. multocida isolates are critical agents in an upper respiratory disease, progressive atrophic rhinitis (PAR). Pasteurella multocida strains associated with PAR usually produce a dermonecrotic toxin, encoded by the toxA gene. The toxA protein of these isolates acts as an essential virulence factor, inducing turbinate atrophy and poor weight gain in pigs. Non-toxigenic P. multocida strains are not usually associated with PAR. Therefore, confirmation of toxin production is important for the diagnosis and control of the disease.

Description:  BactoReal® Kit Pasteurella multocida is a real-time PCR for the qualitative detection of the toxA gene of P. multocida. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Pasteurella multocida was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of Pasteurella multocida toxA DNA by targeting the toxA gene using real-time PCR, supporting diagnosis of progressive atrophic rhinitis and related porcine respiratory diseases.
The toxA gene encodes a dermonecrotic toxin responsible for turbinate atrophy and poor weight gain in pigs. Detecting this gene confirms involvement of toxigenic strains in progressive atrophic rhinitis.
Validated samples are DNA extracts from swabs of the respiratory tract of pigs, processed with appropriate DNA extraction methods.
An internal DNA positive control (IPC) is added during extraction and detected in the Cy5 channel. It monitors DNA extraction efficiency and identifies possible PCR inhibition.
The kit detects at least 40 copies per reaction with a LoD95% of 60 copies. Validation confirmed 100% specificity with no cross-reactions to other tested bacterial species.
