Background:  Porcine hemagglutinating encephalomyelitis virus (PHEV), an RNA virus, is a member of the genus Coronavirus (Betacoronavirus), family Coronaviridae, and highly subclinical prevalent worldwide. Primarily threat to pigs less than three weeks old. This viral disease of piglets is characterized by vomiting, anorexia and neurological symptoms and results either in rapid death or chronic emaciation (vomiting and wasting). The mortality rate in infected piglets under three weeks old is nearly 100 %, but the incidence of infection is small as most piglets are protected by maternal antibodies. Clinically apparent disease develops only when young piglets from non-immune sows are exposed.

Description:  ViroReal® Kit PHEV is a real-time PCR for the qualitative detection of the nucleocapsid protein gene (N gene) of the PHEV. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit PHEV was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the nucleocapsid (N) gene of PHEV RNA and detects amplification via the FAM channel.
PHEV primarily affects piglets less than 3 weeks old. Transmission occurs via oral–nasal routes, and most infections remain subclinical due to maternal antibodies.
The IPC RNA can be added either during RNA extraction for full-process control or directly into the PCR master mix for RT-PCR-only control.
The kit is validated for ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, qTOWER3G, MIC, and cobas z 480.
Primers and probes were validated experimentally and in silico. The kit detects all known PHEV strains and does not cross-react with other coronaviruses, except Bovine Coronavirus.
