Background:  Porcine circovirus type 3 (PCV3) is a novel circovirus species identified in pigs with porcine dermatitis and nephropathy syndrome (PDNS), reproductive failure, respiratory disease and multi-systemic inflammation. Porcine circovirus is a small, non-enveloped DNA virus that contains a single strand of circular DNA. It belongs to the genus Circovirus in the family Circoviridae.

Description:  ViroReal® Kit PCV3 is a real-time PCR for the qualitative detection of the DNA of the capsid gene of porcine circovirus type 3 (PCV3). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit PCV3 was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems) cobas z 480 Analyzer (Roche), and QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit amplifies the capsid gene of PCV3, and detection occurs in the FAM fluorescence channel.
No, the kit is specific for PCV3 and does not cross-react with PCV1 or PCV2.
If not added during extraction, the IPC can be added to the master mix at a 1:100 dilution to monitor for inhibition.
Compatible systems include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Work in dedicated PCR areas, use DNase-free materials and aerosol-resistant pipette tips, and always pipette the positive control last.
