Background:  Porcine cytomegalovirus (PCMV) is a herpesvirus infecting only pigs. PCMV is present worldwide. Most PCMV infections are sub-clinical and clinical disease is rare causing inclusion body rhinitis (IBR), pneumonia, anaemia, fever and abortion or neonatal piglet losses. The virus may also be endemic without causing any apparent clinical disease or economic loss. However, these infected animals can be latent carriers of the virus. In the context of xenotransplantation, PCMV/PRV is the second known zoonotic (disease-inducing) pig virus after hepatitis virus E genotype 3.

Description:  ViroReal® Kit PCMV is a real-time PCR for the qualitative detection of the DNA polymerase (DPOL) gene of the porcine cytomegalovirus (PCMV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

Product Features:
  • Amplification and detection: DPOL gene of PCMV
  • Real-time PCR with rapid hot-start Taq DNA polymerase
  • ROX dye as passive reference
  • Internal Positive Control System to exclude false-negative results
  • Optimized to handle PCR inhibitors
  • PCR platforms: runs on all established standard real-time PCR platforms
  • Harmonized thermal profiles to run RNA and DNA samples simultaneously
  • Available formats: Kit (with reaction mix) or Assay-only

PCR platforms: ViroReal® Kit PCMV was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), and cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit targets the DNA polymerase (DPOL) gene of PCMV, detected via the FAM fluorescence channel.
PCMV is globally distributed and may affect reproductive health in pigs; it also poses risks in xenotransplantation due to its zoonotic potential.
The IPC checks both the quality of DNA extraction and the presence of PCR inhibitors, helping to rule out false-negative results.
Validated systems include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Primers and probes were validated through in silico and experimental testing, confirming detection of all known PCMV strains with no cross-reactivity.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
