Background:  Nocardia is a genus of Gram-positive, facultative intracellular bacteria belonging to the Actinomycetes. Nocardia species are found worldwide in soil, water and plants. This genus has 85 species, of these > 25 have been recognized as causes of human infections, and > 30 are responsible for animal diseases. Nocardia spp. cause opportunistic disease in humans and animals. Nocardiosis is an infectious disease affecting either the lungs, but it may spread to other organ systems, most often the brain and the skin. It may also involve the kidneys, joints, heart, eyes, and bones. Bovine mastitis is the most common clinical manifestation of nocardiosis among domestic ruminants. Nocardial mastitis is observed in dairy herds submitted to inadequate milking management and/or poor hygienic conditions in pre- and post-milking. The most important pathogenic species for humans and animals are N. asteroides complex, N. brasiliensis, N. pseudobrasiliensis, N. otitidiscaviarum, and N. transvalensis.

Description:  BactoReal® Kit Nocardia spp. is a real-time PCR for the qualitative detection of the 16S rDNA gene of Nocardia spp. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Nocardia spp. was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects Nocardia DNA sequences across multiple clinically relevant species including N. asteroides complex and N. brasiliensis.
The kit amplifies the 16S rRNA gene for sensitive and specific detection of Nocardia DNA.
Nocardia DNA is detected in the FAM channel, and the IPC is detected in the Cy5 channel.
