Background:  Mycoplasma hyorhinis causes pneumonia, polyarthritis and polyserositis in swine. Mycoplasma hyorhinis can be isolated e.g. from joints, liver, lung tissues and nasal cavities.

Description:  BactoReal® Kit Mycoplasma hyorhinis is a real-time PCR for the qualitative detection of the rpoB gene of M. hyorhinis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Mycoplasma hyorhinis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit amplifies the rpoB gene of Mycoplasma hyorhinis, allowing species-specific DNA detection by real-time PCR in the FAM channel.
M. hyorhinis is associated with pneumonia, polyarthritis, polyserositis, and has been isolated from central nervous system infections in swine.
Validated materials include DNA extracts from biopsies such as joints, liver, lung tissue, and nasal cavities of pigs.
An internal DNA positive control (IPC), added during DNA extraction and detected in the Cy5 channel, verifies successful DNA extraction and identifies PCR inhibition.
The analytical sensitivity is 10 DNA copies per PCR reaction, with a limit of detection (LoD95%) of 52 copies. Validation showed 100% specificity and no cross-reactions with related bacteria.
