Background:  Mycoplasma bovis belongs to the class Mollicutes and is a cell wall-less bacterium that adheres to host cell surfaces. The organism is typically sensitive to tetracycline, although antimicrobial resistance has increaisngly been reported. The pathogen is causing respiratory disease, otitis media, arthritis, mastitis, keratoconjunctivitis, meningitis and endocarditis in cattle worldwide. It can affect all age groups and all sections.

Description:  BactoReal® Kit Mycoplasma bovis is a real-time PCR for the detection of the rpoB gene of M. bovis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Mycoplasma bovis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects DNA of Mycoplasma bovis by amplifying the rpoB gene using real-time PCR with fluorescence measurement in the FAM channel.
M. bovis is linked to respiratory infections, mastitis, otitis media, arthritis, keratoconjunctivitis, meningitis, and endocarditis in cattle worldwide.
Validated materials include DNA extracts from synovial fluid, bronchoalveolar lavage (BAL), milk, swabs, and biopsies.
The internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, verifies DNA extraction efficiency and identifies possible PCR inhibition.
The kit detects as few as 2 target DNA copies per PCR reaction, with a LoD95% of 4 copies. No cross-reactions were observed with other tested Mycoplasma species.
