Background:  Tuberculosis is caused by members of the Mycobacterium tuberculosis complex (MTBC). The eight closely related species of the MTBC have a wide range of natural hosts, including human hosts (M. tuberculosis, M. africanum, M. canetti), bovine hosts (M. bovis), caprine hosts (M. caprae), rodent hosts (M. microti), and pinned hosts (M. pinnipedii). Bovine tuberculosis remains an important disease in many countries of the world, causing significant economic losses. Wildlife reservois make the eradication difficult and that is why M. bovis has still a high zoonotic potential.

Description:  BactoReal® Kit Mycobacterium tuberculosis complex is a real-time PCR for the qualitatative detection of the ITS2 region of Mycobacterium species belonging to the Mycobacterium tuberculosis complex (M. tuberculosis, M. bovis, M. caprae, M. canetti, M. africanum and M. microti). Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC). To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Mycobacterium tuberculosis complex was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of DNA from the Mycobacterium tuberculosis complex by targeting the ITS2 region using real-time PCR with detection in the FAM channel.
The MTBC includes M. tuberculosis, M. africanum, M. canettii, M. bovis, M. caprae, M. microti, and M. pinnipedii, affecting humans, cattle, goats, rodents, and pinnipeds.
Validated sample types include bronchoalveolar lavage, pleural fluid, nasopharyngeal swabs or aspirates, cerebrospinal fluid, sputum, urine, whole blood, tissue, and paraffin blocks.
An internal DNA positive control (IPC) is added during DNA extraction and detected in the Cy5 channel. It monitors extraction efficiency and identifies PCR inhibition.
The kit has an analytical sensitivity of 5 DNA copies per reaction and high specificity validated across MTBC members, with no cross-reactivity to non-MTBC Mycobacterium species.
