Background:  Mycobacterium avium ssp. paratuberculosis (MAP) causes Johne's disease, a chronic gastroenteritis affecting primarily ruminants and many other species. This disease is one of the most widespread and economically important diseases of ruminants. It has a relatively long incubation period, characterized by the excretion of MAP in faeces for months and years before first clinical symptoms develop.

Description:  BactoReal® Kit Mycobacterium avium ssp. paratuberculosis is a real-time PCR for the qualitative detection of the insertion sequence IS900 of M. avium ssp. paratuberculosis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Mycobacterium avium ssp. paratuberculosis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables detection of Mycobacterium avium ssp. paratuberculosis (MAP) DNA by targeting the IS900 insertion sequence using real-time PCR, with fluorescence measured in the FAM channel.
MAP is the causative agent of Johne’s disease, a chronic gastroenteritis of ruminants that leads to long incubation, progressive weight loss, diarrhoea, and major economic losses in livestock farming.
Validated samples include DNA extracts from faeces, milk, and tissue materials from ruminants and other host species.
An internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, monitors extraction efficiency and identifies potential PCR inhibition.
The analytical sensitivity is 6 DNA copies per PCR with a LoD95% of 43 copies. Validation confirmed 100% specificity with no cross-reactions to other bacteria tested.
