Background:  Lumpy Skin Disease Virus (LSDV) is a dsDNA virus within the genus Capripoxvirus. It is genetically very similar to the other two Capripoxvirus species Goatpox and Sheeppox Virus. There is one serotype of LSDV, which causes the Lumpy Skin Disease in cattle and buffalo. It is transmissed by arthropod vectors. Traditionally, it has been found in southern Africa, but has been steadily spreading north to the Middle East and into eastern Europe and Russia. Goatpox and Sheeppox are viral diseases endemic in Africa, the Middle East, Central Asia, and India, but have recently spread to Eastern Europe, Russia, China, and southeast Asia. The severe clinical symptoms are fever, generalised papules, internal lesions, and death.

Description:  ViroReal® Kit Lumpy Skin Disease Virus is a real-time PCR for the qualitative detection of the DNA of the GPCR gene of the Lumpy skin disease virus (LSDV), Goatpox virus (GTPV) and Sheeppox virus (SPPV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Lumpy Skin Disease Virus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific) and was also tested with LightCycler® 480 II (Roche Diagnostics) and Mx3005P® (Agilent). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the GPCR gene of LSDV, and detection is performed in the FAM channel.
The test may also detect genetically similar viruses such as sheeppox virus and goatpox virus, due to sequence homology.
Use separate workspaces for extraction and amplification, sterile pipette tips, DNAse-free materials, and pipette the positive control last.
Validated systems include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
The kit uses 5′-nuclease assay-based multiplex real-time PCR with fluorescent probes for specific target detection.
