Background:  Listeria monocytogenes causes listeriosis an infectious but not contagious disease affecting primarily domestic animals, especially ruminants (cattle, sheep, goats) and is less common in human beings. Rarely the disease can be found in birds, horses and pigs as well as in rodents and fish. It can also occur in feral animals (e.g. game animals) as well as in poultry and other birds. Listeria are found ubiquitously in soil, water and in poorly made silage.

Description:  BactoReal® Kit Listeria monocytogenes is a real-time PCR for the qualitative detection of the hlyA gene of L. monocytogenes. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Listeria monocytogenes was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of Listeria monocytogenes DNA by targeting the hlyA gene using real-time PCR, supporting diagnosis of listeriosis in animals.
L. monocytogenes is a foodborne pathogen responsible for listeriosis, a disease that can cause sepsis and meningitis, particularly in pregnant women, newborns, elderly, and immunocompromised individuals.
The kit is validated for DNA extracts from cerebrospinal fluid (CSF) and blood.
The kit contains an internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel. It verifies DNA extraction efficiency and rules out PCR inhibition.
The LoD95% is 9 target copies per reaction, with analytical sensitivity down to 1 target copy. No cross-reactivity was observed with other bacteria tested, and all tested L. monocytogenes isolates were correctly identified.
