Background:  Lawsonia intracellularis is a Gram-negative and obligate intracellular bacterium and the causative agent of porcine proliferative enteropathy (PPE), which is a transmissible enteric disease of growing pigs characterized by adenomatous proliferation of immature intestinal epithelial cells in the distal small intestines, particularly in the ileum. The infection is widely distributed among pig herds in many pig-producing countries, and affected pigs may exhibit impaired growth and diarrhoea.

Description:  BactoReal® Kit Lawsonia intracellularis is a real-time PCR for the qualitative detection of the 16S rRNA gene of L. intracellularis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Lawsonia intracellularis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of Lawsonia intracellularis DNA by targeting the 16S rRNA gene using real-time PCR, supporting diagnosis of porcine proliferative enteropathy.
L. intracellularis is the causative agent of porcine proliferative enteropathy (PPE), a widespread enteric disease that leads to impaired growth, diarrhoea, and economic losses in pig production.
The kit is validated for DNA extracts from faeces and biopsies of the intestinal mucosa.
An internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, monitors extraction efficiency and excludes PCR inhibition.
The analytical sensitivity is 10 DNA copies per reaction with a LoD95% of 53 copies per reaction. Validation showed 100% specificity and no cross-reactions with other intestinal bacteria.
