Background:

Kashmir bee virus (KBV) is a positive single-stranded RNA virus within the family Dicistroviridae and the genus Aparavirus. It is closely related to Acute bee paralysis virus (ABPV) and to Israeli acute paralysis virus (IAPV), hence all three are summarized in the so-called AKI-complex. Members of that complex play a role in the sudden collapses of honeybee hives and colonies and cause economical losses worldwide. KBV infects adult bees, that die within days, and brood, but surviving larvae carry the virus latently.

To distinguish between the Aparavirus species within the AKI-complex, ingenetix offers individual real-time PCR Kits for detection of Acute bee paralysis virus, Kashmir bee virus and Israeli acute paralysis virus. Moreover a real-time PCR Kit for the universal detection of aparaviruses is available.

Description:

ViroReal® Kit Kashmir Bee Virus is a real-time PCR for the qualitative detection of the RNA of the capsid protein gene of Kashmir Bee Virus. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms:

ViroReal® Kit Kashmer Bee Virus was validated with the QuantStudio 5 Real-Time PCR System (Thermo Fisher Scientific), and was also tested with QuantStudio 7 Real-Time PCR System (Thermo Fisher Scientific). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. ABI® 7500 Fast Real-Time PCR System (Thermo Fisher Scientific), Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) und cobas z 480 Analyzer (Roche), QuantStudio 6 (Thermo Fisher Scientific)).

Frequently Asked Questions:

The kit targets the capsid protein gene of KBV and detects amplification via the FAM fluorescence channel.
The IPC is added during RNA extraction or, if skipped, diluted 1:100 and added to the PCR master mix to control for inhibition.
Compatible platforms include QuantStudio 5/6/7, qTOWER3G, MIC, and cobas z 480 with FAM and Cy5 detection channels.
Infected adult bees die within days, while larvae may remain latently infected. The virus contributes to colony collapse, especially in presence of other pathogens.
Use separate areas for extraction and amplification, RNase-free materials, and pipette the positive control last.
BactoReal®, MycoReal®, ParoReal® and ViroReal® Kits run with the same thermal cycling conditions.
