Background:  Histophilus somni is an unencapsulated Gram-negative bacterium and a member of the Pasteurellaceae family. It is a common commensal in the lower reproductive and upper respiratory tract of cattle and other ruminants. H. somni is one of the bacterial agents responsible for bovine respiratory disease (BRD) complex and associated with respiratory disease, arthritis, mastitis, myocarditis and reproductive failure in sheep and cattle. Other important bacterial pathogens of BRD are Pasteurella multocida, Mannheimia haemolytica, Trueperella pyogenes and Mycoplasma bovis. Mixed infections with these organisms do occur.

Description:  BactoReal® Kit Histophilus somni is a real-time PCR for the qualitative detection of the 16S rRNA gene of Histophilus somni. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Histophilus somni was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional -  this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit amplifies the 16S rRNA gene of H. somni, with detection in the FAM channel.
Infections can lead to respiratory disease, arthritis, mastitis, myocarditis, and reproductive disorders in cattle and sheep, often in combination with other pathogens such as Pasteurella multocida or Mannheimia haemolytica.
H. somni DNA is detected in the FAM channel, while the IPC is detected in the Cy5 channel.
Strict laboratory workflow separation, sterile consumables, and use of negative extraction controls are recommended to avoid contamination.
Validated systems include ABI 7500, LightCycler® 480 II, Mx3005P®, QuantStudio™, qTOWER3G, Mic instrument, and cobas z 480.
