Background:  Glaesserella parasuis (former Haemophilus parasuis) is a Gram-negative bacterium (member of the Pasteurellaceae) of the upper respiratory tract of swine. It is the aetiological agent of Glässers disease, a severe systemic disease characterized by fibrinous polyserositis, pericarditis, arthritis and meningitis. In addition to the acute form of the disease, Glaesserella parasuis is an important component of the Porcine Respiratory Disease Complex (PRDC). Fifteen serovars of Glaesserella parasuis have been reported, but a high percentage of the evaluated isolates are non typeable. Isolates from the upper respiratory tract are mainly apathogenic strains of clinically healthy swine.

Description:  BactoReal® Kit Glaesserella parasuis is a real-time PCR for the qualitative detection of the 16S rRNA gene of Glaesserella parasuis (former H. parasuis). Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Glaesserella parasuis was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

The kit enables molecular detection of Glaesserella parasuis DNA by targeting the 16S rRNA gene using real-time PCR, supporting diagnosis of Glässer’s disease and porcine respiratory disease complex (PRDC).
G. parasuis causes Glässer’s disease, a systemic condition characterized by fibrinous polyserositis, pericarditis, arthritis, and meningitis. It also plays a role in PRDC, leading to economic losses in pig production.
Validated samples include DNA extracts from synovial, pleural, pericardial, peritoneal, and cerebrospinal fluids.
The kit contains an internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, to monitor DNA extraction efficiency and exclude PCR inhibition.
The analytical sensitivity is 10 target copies per reaction, with a limit of detection (LoD95%) of 72 copies. Cross-reactivity was noted with some Actinobacillus and Pasteurella species, but specificity against other respiratory bacteria was confirmed.
