Background:  Escherichia coli is a Gram-negative bacterium that is a member of the normal gut flora of humans and animals. Most E. coli strains are harmless, but some serotypes carry virulence factors, which enable them to be pathogenic in target hosts. These pathogenic E. coli are implicated in a number of diseases including diarrhoea, septicaemia and urinary tract infections in humans and animals. The dxs gene (1-deoxyxylulose-5-phosphate synthase gene) can be found in E. coli and Shigella strains.

Description:  BactoReal® Kit Escherichia coli is a real-time PCR for the qualitative detection of the dxs gene of E. coli and Shigella. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Escherichia coli was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects Escherichia coli DNA by amplifying the dxs gene using real-time PCR with fluorescence measurement in the FAM channel.
E. coli is a common gut commensal, but pathogenic strains are linked to diarrhoea, pneumonia, septicaemia, and urinary tract infections in humans and animals.
The kit is validated for DNA extracted from blood, tissue, and milk samples.
An internal DNA positive control (IPC) is added during DNA extraction and detected in the Cy5 channel, ensuring extraction efficiency and ruling out PCR inhibition.
The detection limit is 1000 copies per reaction. The kit detects all known E. coli strains but may show cross-reaction with Shigella species.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
