Background:  Encephalomyocarditis virus (EMCV) is one of three virus species in the genus Cardiovirus in the family Picornaviridae and is a single stranded RNA (ssRNA) virus that causes encephalomyocarditis and reproductive disease in swine and zoologic mammals. Infected piglets suffer from encephalitis, myocarditis and sudden death. Reproductive problems include poor conception rates, embryo resorption, mummification, stillbirths, abortions and neonatal death. The disease affects the nervous, reproductive, respiratory and circulatory system of pigs. Fatal myocarditis due to EMC virus infection has been seen in primates, even-and odd-toed ungulates, elephants, carnivores and rodents. The disease can be found worldwide. The most important mode of virus transmission seems to be rodent-to-pig transmission, and rodents act as the major reservoir of the disease. Rats and other rodents probably infect pigs directly or through diseased carcasses and contaminated feeds or water supplies.

Description:  ViroReal® Kit EMCV is a real-time PCR for the qualitative detection of the 3´UTR RNA of EMCV. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit EMCV was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the highly conserved 3′ untranslated region (3′ UTR) of EMCV RNA, detected via the FAM channel.
Rodents are considered the primary reservoir and may transmit EMCV to pigs through direct contact, carcasses, or contaminated feed and water.
The RNA IPC confirms extraction efficiency and RT-PCR performance, ensuring the validity of negative test results.
Validated systems include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Primers and probes were validated in silico and experimentally to confirm detection of known EMCV strains and avoid cross-reactivity.
