Background:  Coxiella burnetii (C. burnetii) is a Gram-negative, obligate intracellular, zoonotic bacterial pathogen, which is the causative agent of Q fever (Coxiellosis). The disease is spread through inhalation of a spore-like cell variant or close contact with body fluids. Cattle, sheep and goats represent the main animal reservoir. Seropositivity of ruminants for C. burnetii is associated with abortion, stillbirth and fertility problems. The high resistance to standard disinfectants and environmental influences renders disease control difficult.

Description:  BactoReal® Kit Coxiella burnetii is a real-time PCR for the qualitative detection of the IS1111 insertion sequence of Coxiella burnetii. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Coxiella burnetii was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit amplifies the IS1111 insertion sequence of Coxiella burnetii, detected in the FAM channel.
In animals, C. burnetii infection can cause abortion, stillbirth, and reduced fertility, while in humans it causes Q fever, characterized by fever, pneumonia, or hepatitis.
C. burnetii DNA is detected in the FAM channel, and the IPC is detected in the Cy5 channel.
The IPC excludes false negatives by verifying DNA extraction and ruling out PCR inhibition.
The kit was tested against Legionella pneumophila and Rickettsia helvetica; no cross-reactions were observed.
