Background:  Clostridium difficile is a Gram-positive spore-forming anaerobic bacterium, commonly found in low concentrations in the intestine of clinically normal animals and humans. In higher concentration Clostridium difficile is an important cause of diarrhea in neonatal swine. Affected piglets may have dyspnea, abdominal distention, and scrotal edema. Diarrhea may not be present in all pigs affected. Pathogenic C. difficile strains produce multiple toxins. The best characterized are enterotoxin (Clostridium difficile toxin A causing fluid secretion into the gut lumen) and cytotoxin (Clostridium difficile toxin B), both of which may produce diarrhea and inflammation.

Description:  BactoReal® Kit Clostridium difficile is a real-time PCR for the qualitative detection of the Clostridium difficile toxin A (tcdA) gene and Clostridium difficile toxin B (tcdB) gene of C. difficile. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Clostridium difficile was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional -  this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit amplifies the tcdB gene detected in the FAM channel and the tcdA gene detected in the VIC channel.
C. difficile is a leading cause of neonatal diarrhea in piglets and may also cause edema, distention, and other gastrointestinal symptoms.
Toxin B is detected in the FAM channel, toxin A in the VIC channel, and the IPC in the Cy5 channel.
Positive and negative controls along with the IPC confirm the validity of test results.
The limit of detection was determined as 33 copies per reaction for toxin B and 52 copies per reaction for toxin A.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
