Background:
Chronic bee paralysis virus (CBPV) is an enveloped virus with a genome consisting of three single plus-stranded RNA
molecules. It commonly affects the honeybee Apis mellifera. The unclassified virus is phylogenetically placed between
the Nodaviridae (insect and fish viruses) and Tombusviridae (plant viruses) family. It is suggested that CBPV is the
prototype of a new family of positive single-stranded RNA viruses. Infections were rather unusual but a continuous
increase of incidents worldwide was reported recently. The very characteristic symptoms like inability to fly, trembling,
ataxia, and shiny hairless bodies could even lead to collapses of the bee colonies. Infected symptomatic bees, mostly
adults, die within a few days.
Description:
ViroReal® Kit Chronic Bee Paralysis Virus is a real-time PCR for the qualitative detection of the RNA of the RdRp gene of chronic bee paralysis virus. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms:
ViroReal® Chronic Bee Paralysis Virus was validated with the QuantStudio™ 5 Real-Time PCR System (Thermo Fisher Scientific), and was also tested with ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), and cobas z 480 Analyzer (Roche)).

The kit is designed for qualitative detection of RNA from Chronic Bee Paralysis Virus (CBPV) by targeting the RdRp gene using real-time RT-PCR.
CBPV causes trembling, inability to fly, ataxia, and hairless shiny abdomens. Infected adult bees often die within a few days, and outbreaks may lead to colony collapse.
The kit includes an artificial RNA internal positive control (IPC) added during extraction to monitor RNA integrity and PCR inhibition, detected in the Cy5 channel.
The limit of detection (LoD95%) is approximately 2.5 RNA copies per reaction.
The kit is validated for RNA extracted from honeybee colony material, including homogenized bee samples.
