Background:  The CSF (classical swine fever) virus is a single strain, positive-sense ((+)sRNA) RNA virus belonging to the genus Pestivirus in the family Flaviviridae. The CSF virus is the infectious agent of classical swine fever (CSF) or hog cholera (also sometimes called pig plague based on the German word “Schweinepest”), which is a highly contagious disease of swine. Swine fever causes fever, skin lesions, convulsions, and usually death. The effect of different CSF virus strains varies widely, leading to a wide range of clinical signs. There is only one serotype of the virus and attenuated vaccines are available. Since the virus is classified as notifiable disease at the World Organisation for Animal Health (WOAH) an outbreak could significant effect all swine trades and production. This virus is closely related to ruminant pestiviruses that cause bovine viral diarrhoea (BVDV) and border disease (BDV) of sheep, both of which can also infect pigs naturally causing reproductive problems.

Description:  ViroReal® Kit CSF Virus is a real-time PCR for the qualitative detection of the 5´UTR of the CSF virus genome. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit CSF Virus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific), qTOWER3G (Analytik Jena), Mic instrument (bio molecular systems), and cobas z 480 Analyzer (Roche)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit amplifies the 5′UTR of CSFV RNA and detects fluorescence in the FAM channel.
The IPC helps confirm successful RNA extraction and checks for reverse transcription or PCR inhibition, reducing false-negative results.
Validated instruments include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Yes, the IPC RNA can be added during extraction or directly into the master mix to control RT-PCR only.
The kit cross-reacts with BVDV and BDV, both of which can naturally infect pigs and cause reproductive disorders.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
