Background:  Brucellosis - also called Bang's disease, Crimean fever, Gibraltar fever, Malta fever, Maltese fever, Mediterranean fever, rock fever, or undulant fever - is a zoonotic infection of domesticated and wild animals, caused by bacteria of the genus Brucella. Species infecting domestic livestock are B. melitensis (goats and sheep), B. suis (pigs), B. abortus (cattle and bison), B. ovis (sheep) and B. canis (dogs). Humans become infected by ingestion of food products of animal origin (such as undercooked meat or unpasteurized milk or dairy products), direct contact with infected animals, or inhalation of infectious aerosols. Transmission from human to human is rare but possible.

Description:  BactoReal® Kit Brucella spp. is a real-time PCR for the qualitative detection of th IS711 region of B. abortus, B. ovis, B. melitensis, B. suis and B. canis. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Brucella spp. was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects DNA of Brucella species by amplifying the IS711 insertion sequence, with specific signals measured in the FAM channel.
The kit detects B. abortus, B. melitensis, B. suis, B. ovis, and B. canis, all of which are associated with brucellosis in livestock and companion animals.
Brucellosis can cause reproductive disorders in animals such as abortions, infertility, and reduced milk production. In humans it manifests as undulant fever and systemic infection.
Validated samples include DNA extracts from blood, milk, lymph tissue, bone marrow, and biopsies.
The kit includes an internal DNA positive control (IPC), added during DNA extraction and detected in the Cy5 channel, to monitor extraction efficiency and detect PCR inhibition.
