Background:  Brachyspira pilosicoli (previously called Serpulina pilosicoli) is an intestinal spirochaete. This bacterium colonizes the large intestine of various species, including humans. It is the etiologic agent of human and animal intestinal spirochetosis and rarely implicated as a cause of bacteremia. For example, it is the causative agent of porcine colonic spirochaetosis (PCS). Brachyspira pilosicoli is an anaerobic bacterium but is aerotolerant due, at least in part, to high NADH oxidase activity.

Description:  BactoReal® Kit Brachyspira pilosicoli is a real-time PCR for the qualitative detection of the NADH oxidase (nox) gene of B. pilosicoli. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Brachyspira pilosicoli was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables molecular detection of Brachyspira pilosicoli DNA by targeting the nox gene using real-time PCR, supporting diagnosis of intestinal spirochetosis in animals.
B. pilosicoli colonizes the large intestine of humans and animals and is the causative agent of porcine colonic spirochetosis and intestinal spirochetosis in other hosts. It may occasionally cause bacteremia in humans.
Validated samples include DNA extracts from faeces and intestinal biopsies.
The internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, monitors DNA extraction efficiency and excludes PCR inhibition.
The analytical sensitivity is 10 target copies per PCR reaction with a limit of detection (LoD95%) of 25 copies. Validation confirmed 100% specificity with no cross-reactions against other intestinal bacteria.
