Background:  Neboviruses are non-enveloped, single-stranded, linear, positive-sense RNA viruses ((+)ssRNA), belonging to the Caliciviridae family. The only currently known species in the genus Nebovirus is Newbury-1 virus. Cattle, especially calves, serve as natural hosts. Neboviral infection is associated with gastrointestinal disorders and necrotizing hepatitis leading to fatal hemorrhages in calves. The disease often remains endemic due to the lack of a commercially available vaccine.

Description:  ViroReal® Kit Nebovirus is a real-time PCR for the qualitative detection of the RNA of ORF 1 of Bovine Nebovirus. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Bovine Nebovirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas z 480 Analyzer (Roche) und QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the ORF1 region of BNeV RNA, detected in the FAM channel.
BNeV spreads among calves via the fecal–oral route, leading to outbreaks of diarrhea and enteric disease in herds.
The RNA IPC confirms successful RNA extraction and rules out RT-PCR inhibition, thereby reducing the chance of false-negative results.
The kit is validated for ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, MIC, qTOWER3G, and cobas z 480.
Primers and probes were validated in silico and experimentally, confirming detection of all known BNeV strains without cross-reactivity.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
