Background:  Bluetongue virus (BTV) is a segmented, linear, double-stranded RNA virus (dsRNA), belonging to the Reoviridae family and is divided into 28 serotypes. The virus is transmitted by biting midges and infects wild and domestic ruminants such as sheep and less frequently goats, deer and cattle. There is no appropriate treatment of acute disease, which is associated with high morbidity and mortality in some sheep breeds. Prophylactic immunisation with attenuated and inactivated vaccines is available. However, they do not cover all virulent BTV serotypes.

Description:  ViroReal® Kit Bluetongue Virus (BTV) is a real-time PCR for the qualitative detection of the segment 10 (NS3) of BTV. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Bluetongue Virus (BTV) was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas Z 480 Analyzer (Roche) QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables qualitative detection of RNA from Bluetongue Virus (BTV) by targeting the segment 10 (NS3) region using reverse transcription real-time PCR.
BTV causes a severe hemorrhagic disease in ruminants, particularly sheep, and is transmitted by biting midges. Infections can lead to high morbidity and mortality in susceptible breeds.
An artificial RNA internal positive control (IPC) is added during RNA extraction to monitor extraction efficiency and detect RT-PCR inhibition. It is detected in the Cy5 channel.
Validated sample materials include RNA extracted from spleen, lymph nodes, and blood of ruminants.
The analytical sensitivity is at least 50 target copies per reaction. Linearity is confirmed between 100 and 1,000,000 copies/reaction.
