Background:  Actinobacillus pleuropneumoniae (APP) causes respiratory disease in swine (pig haemorrhagic pleuropneumonia, actinobacillosis). It is a Gram-negative coccobacillus and a pathogen of the respiratory tract. Clinical signs of the acute disease are dyspnea, coughing, anorexia, depression, fever and sometimes vomiting. The disease can progress very rapidly and death can occur within a few hours. Chronic infections are characterized by cough and pleuritis. Many herds are infected with APP without presenting any clinical evidence of the disease. Carrier pigs harbour APP in their nasal cavities and/or tonsils.

Description:  BactoReal® Kit Actinobacillus pleuropneumoniae is a real-time PCR for the qualitative detection of the apxIVA gene of Actinobacillus pleuropneumoniae. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

  • Real-time PCR with rapid hot-start Taq DNA polymerase
  • ROX™ dye as passive reference
  • Internal Positive Control System to exclude false-negative results
  • Optimized to handle PCR inhibitors
  • PCR platforms: runs on all established standard real-time PCR platforms
  • Harmonized thermal profiles to run RNA and DNA samples simultaneously
  • Available formats: Kit (with reaction mix) or Assay-only

PCR platforms: BactoReal® Kit Actinobacillus pleuropneumoniae was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

Frequently Asked Questions:

The kit detects DNA of Actinobacillus pleuropneumoniae by targeting the apxIVA gene using real-time PCR with FAM channel detection.
A. pleuropneumoniae is the causative agent of pig haemorrhagic pleuropneumonia, leading to acute respiratory disease with high mortality, and chronic infections characterized by cough and pleuritis.
The kit is validated for DNA extracts from tissues such as lung and tonsil of swine.
An internal DNA positive control (IPC), added during extraction and detected in the Cy5 channel, ensures efficient DNA extraction and identifies possible PCR inhibition.
The kit detects as few as 10 DNA copies per PCR reaction with a LoD95% of 18 copies. No cross-reactivity was observed with tested bacteria including H. influenzae, P. multocida, M. hyopneumoniae, and L. monocytogenes.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
