Background:  Porcine epidemic diarrhea virus (PEDV), a member of the genus Coronavirus (Alphacoronavirus), family Coronaviridae, is a positive-sense, enveloped, single-stranded RNA virus. Severity of disease is variable and dependent on the epidemiological status of the herd. When epidemic, PEDV causes acute watery diarrhea and vomiting in a large proportion at all ages of swine. If endemic, then diarrhea is observed with lower morbidity but higher mortality rate in suckling and recently weaned pigs. Infected sows and gilts show less reproduction performance. The PED virus is clinically similar to, but antigenically distinct from transmissible gastroenteritis virus (TGEV).

Swine delta coronavirus (SDCV, also called porcine delta coronavirus PDCoV), a member of the genus Coronavirus (Deltacoronavirus), family Coronaviridae, is a positive-sense, enveloped, single-stranded RNA virus. SDCV is a new virus first found in pigs in Hong Kong in 2012 and 2014 in the United States. Swine delta coronavirus causes acute watery diarrhea and vomiting in a large proportion at all ages of swine. Swine delta coronavirus infection is clinically similar to, but genetically distinct from porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV).

Description:  ViroReal® Kit PEDV & SDCV is a real-time PCR for the qualitative detection of the nucleocapsid protein gene (N gene) of PEDV (porcine epidemic diarrhea virus) and of the 3’UTR of SDCV (swine delta coronavirus, PDCoV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

Frequently Asked Questions:

The test detects PEDV in the FAM channel and SDCV in the VIC channel using specific probes, allowing simultaneous differentiation.
Both viruses cause acute watery diarrhea and vomiting in pigs of all ages, with high mortality especially in neonatal piglets.
The IPC confirms the effectiveness of RNA extraction and absence of RT-PCR inhibition, minimizing false-negative results.
Validated platforms include ABI 7500, LightCycler 480 II, Mx3005P, QuantStudio 5/6/7, qTOWER3G, MIC, and cobas z 480.
Primers and probes were validated in silico and experimentally, showing no cross-reactivity with TGEV, PHEV, PRRSV, PPV, or PCV2.
BactoReal® , MycoReal® , ParoReal ® and ViroReal® Kits run with the same thermal cycling conditions.
