Background:  Trichinellosis is a food-borne parasitic zoonosis caused by nematodes of the genus Trichinella. Twelve genotypes of Trichinella have been identified worldwide, four of which are confirmed to exist in Europe: T. spiralis, T. nativa, T. britovi and T. pseudospiralis. Two main clades are recognized in the genus: one group (T. britovi, T. murrelli, T. nativa, T. nelsoni, T. spiralis) that encapsulates in host muscle tissue and a second (T. papuae, T. pseudospiralis, T. zimbabwensisi) that does not.

Description:  ParoReal® Kit Trichinella spiralis & britovi is a real-time PCR for the qualitative detection of the internal transcribed spacer of Trichinella spiralis and T. britovi. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ParoReal® Kit Trichinella spiralis & britovi was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit enables qualitative detection of Trichinella spiralis and Trichinella britovi DNA by amplifying internal transcribed spacer regions with real-time PCR.
These parasites cause trichinellosis, a zoonotic disease transmitted through consumption of raw or undercooked meat containing infective larvae.
The kit contains an artificial DNA internal positive control (IPC) to monitor DNA extraction and detect possible PCR inhibition.
The kit is validated for DNA extracted from meat samples.
The detection limit is approximately 5–10 target DNA copies per PCR reaction.
