Background:  The Gram-negative bacterium Pasteurella multocida is able to cause disease in a wide range of animal species and in humans. Pasteurella multocida can cause fowl cholera disease in poultry, hemorrhagic septicemia and shipping fever in cattle, atrophic rhinitis in pigs and is the most common cause of respiratory disease, primarily rhinitis, in rabbits. Although Pasteurella multocida is part of the flora in the respiratory tract of pigs, the bacterium can induce hemorrhagic septicemia or, usually as a secondary pathogen invading lungs injured by other bacteria or viruses, pneumonia. A subset of Pasteurella multocida isolates are critical agents in an upper respiratory disease, progressive atrophic rhinitis (PAR) in pigs.

Description:  BactoReal® Kit Pasteurella multocida uses real-time PCR for the detection of the 16S rRNA gene of P. multocida (subsp. multocida, gallicida, septica and tigris). Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC). To minimize PCR cross-contamination,the reaction mix included contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Pasteurella multocida is developed and validated for the Applied Biosystems® 7500 System (Thermo Fisher Scientific), LightCycler® 480 (Roche) and Mx3005P® QPCR System (Agilent), but is also suitable for other real-time PCR instruments.

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit detects DNA of Pasteurella multocida by targeting the 16S rDNA gene using real-time PCR with FAM channel detection.
P. multocida can cause fowl cholera in poultry, hemorrhagic septicemia and shipping fever in cattle, atrophic rhinitis in pigs, and respiratory infections in rabbits. It may also act as a secondary pathogen in pneumonia.
The kit is validated for DNA extracts from blood, swabs, and biopsies of different animal species.
An internal DNA positive control (IPC), added during DNA extraction and detected in the Cy5 channel, ensures DNA extraction efficiency and detects possible PCR inhibition.
The kit detects as few as 2 DNA copies per PCR reaction. Specificity testing confirmed high selectivity for P. multocida subspecies without cross-reactivity to related bacteria such as Haemophilus or Mannheimia.
