Background:  Streptococcus dysgalactiae is a Gram-positive bacterium belonging to the group C streptococci. It is known to cause clinically acute and subclinical mastitis in cows, ewes and goats, as well as polyarthritis.

Description:  BactoReal® Kit Streptococcus dysgalactiae is a real-time PCR for the qualitative detection of S. dysgalactiae (S. dysgalactiae subsp. equisimilis and S. dysgalactiae subsp. dysgalactiae). This test is performed in a multiplex real-time PCR format to enable a differentiation between S. dysgalactiae from other Streptococcus species, especially S. canis. It is based on the amplification and detection of the 23S rRNA gene and 16S rRNA gene. In FAM channel the 23S rRNA gene of S. dysgalactiae and S. canis is detected. In VIC/HEX channel the 16S rRNA gene of several Streptococcus species (S. dysgalactiae, S. agalactiae, S. phocae, S. porcinus, S. didelphis) except that of S. canis is detected. Thus, samples showing a positive signal in FAM as well as in VIC/HEX channel are positive for S. dysgalactiae. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Streptococcus dysgalactiae was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the 23S rRNA gene of S. dysgalactiae and S. canis, and the 16S rRNA region of several Streptococcus species (except S. canis). Signals are measured in FAM for S. dysgalactiae/S. canis and VIC for other Streptococcus spp.
S. dysgalactiae is a group C streptococcus that causes acute and subclinical mastitis in cows, ewes, and goats, and may also lead to polyarthritis in livestock.
The kit is validated for DNA extracts from milk and other veterinary sample materials appropriate for mastitis diagnostics.
An internal DNA positive control (IPC), detected in the Cy5 channel, is spiked into each sample during DNA extraction to confirm extraction efficiency and exclude inhibition.
The analytical sensitivity is 5 DNA copies per reaction, with a limit of detection (LoD95%) of 32 copies. Specificity was confirmed, with weak cross-reactions only observed in FAM channel with S. intermedius, S. pyogenes and S. constellatus.
