Background:  Streptococcus agalactiae (also known as Group B streptococcus or GBS) is a beta-hemolytic Gram-positive streptococcus. It is pathogenic primarily for animals, but can also infect humans. In cattle, it causes mastitis, which leads to diminished milk production. Transmission occurs during milking. Streptococcus agalactiae secretes a protein (CAMP factor) which causes haemolysis in the presence of beta-toxin of Staphylococcus aureus in blood agar.

Description:  BactoReal® Kit Streptococcus agalactiae is a real-time PCR for the qualitative detection of the cfb gene of S. agalactiae. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: BactoReal® Kit Streptococcus agalactiae was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The kit targets the cfb gene of Streptococcus agalactiae using real-time PCR with fluorescence detection in the FAM channel.
S. agalactiae, also known as Group B streptococcus (GBS), causes mastitis in cattle, leading to reduced milk production and economic losses. It may also affect other animals and occasionally humans.
The kit has been validated for DNA extracts from milk and other veterinary specimens suitable for detecting S. agalactiae.
An internal DNA positive control (IPC) is included, added during DNA extraction and detected in the Cy5 channel. It verifies DNA extraction and detects PCR inhibition.
The kit shows an analytical sensitivity of 1 DNA copy per reaction with a LoD95% of 3 copies. Validation confirmed 100% specificity with no cross-reactions to other bacteria.
