Background:  The caprine arthritis - encephalitis virus (CAEV) of goats is an enveloped, single-stranded RNA small ruminant lentivirus (SRLV) in the family Retroviridae. There are several, genetically distinct isolates of the virus that differ in virulence. The CAE-virus is closely related to the Maedi-Visna virus (MVV) of sheep. Both infect cells of the monocyte/macrophage lineage and cause lifelong infections. Transmission of the CAEV is through ingestion of virus-infected goat milk but also through virus-infected secretion of affected animals and droplet infection. Clinical symptoms such as severe arthritis of the carpal joint are seen in approximately 20 % of infected goats. Subclinical or clinical interstitial pneumonia, mastitis with lower milk production and chronic wasting have also been attributed to infection with the virus.

Description:  ViroReal® Kit CAEV is a real-time PCR for the qualitative detection of the RNA of the gag polyprotein gene of Caprine Arthritis and Encephalitis Virus (CAEV). Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit CAEV was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas z 480 Analyzer (Roche), and QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The assay detects RNA of Caprine Arthritis Encephalitis Virus (CAEV) by targeting the gag polyprotein gene using reverse transcription real-time PCR.
The assay is validated for RNA extracted from blood mononuclear cells (PBMC).
The artificial RNA IPC is added during extraction and detected in the Cy5 channel to verify RNA extraction and check for PCR inhibition.
The assay detects at least 100 RNA target copies per PCR reaction and shows linearity from 100 to 10,000,000 copies.
The assay includes detection reagents only, while the kit contains a full reagent set including PCR mix and water.
