Background:  Neboviruses are non-enveloped, single-stranded, linear, positive-sense RNA viruses ((+)ssRNA), belonging to the Caliciviridae family. The only currently known species in the genus Nebovirus is Newbury-1 virus. Cattle, especially calves, serve as natural hosts. Neboviral infection is associated with gastrointestinal disorders and necrotizing hepatitis leading to fatal hemorrhages in calves. The disease often remains endemic due to the lack of a commercially available vaccine.

Description:  ViroReal® Kit Nebovirus is a real-time PCR for the qualitative detection of the RNA of ORF 1 of Bovine Nebovirus. Our carefully designed primers and probes ensure the highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Bovine Nebovirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent) and LightCycler® 480 II (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), cobas z 480 Analyzer (Roche), and QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

Assay: Optional - this product is also available as an assay-only format with primers, probe and a positive control, but without reaction mix.

The assay qualitatively detects RNA of Bovine Nebovirus (BNeV) by targeting the ORF1 gene using reverse transcription real-time PCR.
The assay is validated for RNA extracted from bovine fecal samples.
The IPC monitors RNA extraction efficiency and detects possible RT-PCR inhibition. It is added during extraction and detected in the Cy5 channel.
The analytical sensitivity is at least 10 RNA copies per reaction, with linearity from 100 to 1,000,000 copies per reaction.
The assay contains only detection reagents and requires an external PCR mix, while the kit includes a complete reagent set including PCR mix and water.
