Background:  Rotavirus is a segmented double-stranded (ds) RNA virus of the family Reoviridae and is classified into at least 7 groups (A to G). This virus is the causative agent of severe diarrhoea in almost every mammal species (especially cattle, pig, dog, cat horse and humans) and birds. Rotaviruses are thought to be mostly species specific, but some transmission between species occurs. Rotaviruses of group A–C infect both humans and animals, while rotaviruses of group D–G infect only animals. Group A is the most prevalent one and causes approximately 90% of infections in animals and humans. Furthermore, pigs can be sporadically also infected by group B, C and E. Cattles are sporadically also infected by group B and C. Group D, F and G are found in avians.

Description:  ViroReal® Kit Rotavirus is a real-time PCR for the qualitative detection of the non-structural protein 4 (NSP4) gene of human rotavirus genotype A, of most strains of genotype B and C, and of some strains of the other genotypes D, E, F and G. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Rotavirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Rotaviruses are the most common cause of severe diarrhoea in infants and young children worldwide and can also infect animals. Rapid detection supports early diagnosis and prevention of outbreaks.
Validated specimens are RNA extracts from faecal samples, the standard clinical material for rotavirus testing.
An internal RNA positive control (IPC) is added during RNA extraction and detected in the Cy5 channel. It monitors RNA extraction efficiency and rules out PCR inhibition.
The limit of detection (LoD95%) is 115 RNA copies per reaction. The kit showed 100% specificity with no cross-reactions in validation tests and linear detection between 100 and 1,000,000 copies per reaction.
