Background:  Influenza is an acute infectious disease caused by influenza virus A, B or, to a much lesser extent, influenza virus C. Influenza viruses are enveloped viruses with single-stranded, segmented RNA with negative polarity as genome. These viruses can be found worldwide. Epidemics and pandemics are mainly caused by influenza virus A, due to antigenic drift of the hemagglutinin and neuraminidase molecules. Type B and C influenza viruses are isolated almost exclusively from humans, while influenza A viruses infect a wide variety of warm-blooded animals.

Description:  ViroReal® Kit Influenza A/B is a real-time PCR for the qualitative detection of RNA of the matrix protein gene of influenza A virus and the hemagglutinin gene of influenza B virus. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen, as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Influenza A/B was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

Frequently Asked Questions:

The kit detects RNA of the matrix protein gene of influenza A virus in the FAM channel and the hemagglutinin gene of influenza B virus in the VIC channel using multiplex one-step RT-PCR.
Influenza A and B viruses are major causes of seasonal epidemics and severe respiratory illness. Early molecular detection allows timely antiviral therapy and infection control measures.
Validated samples are RNA extracts from human upper respiratory tract specimens, including nasal, nasopharyngeal, and oropharyngeal swabs.
An internal RNA positive control (IPC) is included, added during extraction, and detected in the Cy5 channel. It monitors extraction efficiency and excludes false negatives due to inhibition.
The LoD95% is 5.7 target copies per reaction for influenza A and 6.0 target copies per reaction for influenza B. Diagnostic evaluation showed 100% sensitivity and 100% specificity.
