Background:  Adenoviruses are non-enveloped, double-stranded, linear DNA viruses (dsDNA) belonging to the family Adenoviridae, genus Mastadenovirus. At present seven human adenovirus species are known (A-G) which are divided in more than 50 serotypes. They can cause diseases of the respiratory tract, but also gastroenteritis, conjunctivitis, cystitis, rhinitis, pharyngitis or diarrhoea. Human adenoviruses have a worldwide distribution.

Description:  ViroReal® Kit Adenovirus is a real-time PCR for the qualitative detection of the hexon gene of the so far known species A-G of adenovirus. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Adenovirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

The kit detects the hexon gene of adenoviruses belonging to species A–G, covering all currently known clinically relevant human adenoviruses.
Adenoviruses are associated with respiratory tract infections, gastroenteritis, conjunctivitis, and systemic disease. Rapid molecular detection supports early diagnosis and appropriate patient management.
Validated specimens are DNA extracts from respiratory tract samples, including nasopharyngeal swabs and aspirates, and cerebrospinal fluid.
The kit contains an internal DNA positive control (IPC) detected in the Cy5 channel. It is added during DNA extraction and monitors both extraction efficiency and potential PCR inhibition.
The LoD95% is 137 copies/reaction for adenovirus B, 120 copies/reaction for adenovirus C, and 40 copies/reaction for adenovirus G. Diagnostic evaluation showed 100% sensitivity and 100% specificity in tested samples.
