Background:  Human parechoviruses are non-enveloped, positive single-stranded RNA viruses (ss(+)RNA) and are members of the Picornaviridae family. Six serotypes of human parechovirus have been identified (human parechovirus 1-6). Parechoviruses are responsible for gastrointestinal, respiratory and central nervous system infections. 95% of humans are carriers of human parechovirus since early life.

Description:  ViroReal® Kit Parechovirus is a real-time PCR for the qualitative detection of the polyprotein gene of human parechovirus strains 1-6. Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Parechovirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio™ 5 / 6 / 7 (Thermo Fisher Scientific)).

The kit detects RNA of the polyprotein gene of Human Parechovirus using one-step reverse transcription real-time PCR with probe detection in the FAM channel.
Human parechoviruses are linked to gastrointestinal, respiratory, and central nervous system infections. Severe disease may occur in neonates and immunocompromised individuals.
Validated samples include RNA extracts from serum, cerebrospinal fluid, nasal discharge, urine, and stool.
An internal RNA positive control (IPC) is added during sample extraction and detected in the Cy5 channel, ensuring RNA extraction efficiency and detecting potential RT-PCR inhibition.
The analytical sensitivity is approximately 100 RNA copies per PCR, with linear detection from 100 to 1,000,000 copies per reaction.
