Background:  Human parainfluenzaviruses (HPIV) are enveloped, negative single-stranded RNA viruses (ss(-)RNA) of the family Paramyxoviridae. At present four serotypes are known (types 1, 2, 3, 4). Parainfluenzavirus type 1 and 3 belong to the genus Respirovirus, while parainfluenzavirus type 2 and 4 belong to the genus Rubulavirus. Human parainfluenzavirus type 1 (HPIV-1) is the major cause of acute croup, but can also cause respiratory tract infections at all ages. Human parainfluenzavirus type 2 (HPIV-2) can also cause croup as well as upper and lower respiratory tract illnesses. Human parainfluenzavirus type 3 (HPIV-3) causes bronchiolitis and pneumonia. Human parainfluenzavirus type 4 with the subtypes 4a and 4b is less likely to cause severe disease.

Description:  ViroReal® Kit Parainfluenzavirus uses RT real-time PCR for the detection of the hemagglutinin-neuraminidase glycoprotein gene (HN gene) of human parainfluenzavirus types 1, 2, 3 and 4 (HPIV-1, HPIV-2, HPIV-3, HPIV-4). Our carefully designed primers and probe ensure highest sensitivity and specificity. The kit consists of an assay mix for the detection of the pathogen as well as a positive control and an internal positive control (IPC) system. To minimize PCR cross-contamination, the included reaction mix contains dUTP and uracil-N glycosylase (UNG).

PCR platforms: ViroReal® Kit Parainfluenzavirus was validated with the ABI® 7500 Fast Real-Time PCR System (fast cycle parameters are not supported, Thermo Fisher Scientific), and was also tested with Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche) and cobas Z 480 Analyzer (Roche). It is also compatible with other real-time PCR instruments which detect and differentiate fluorescence in the FAM and Cy5 channel (e.g. QuantStudio 5 / 6 / 7 (Thermo Fisher Scientific)).

The kit detects RNA of the hemagglutinin-neuraminidase (HN) gene of human parainfluenzavirus types 1–4. HPIV-1 and HPIV-3 are detected in the FAM channel, while HPIV-2 and HPIV-4 are detected in the VIC channel.
HPIVs are major causes of respiratory illnesses. HPIV-1 and HPIV-2 can cause croup, HPIV-3 is associated with bronchiolitis and pneumonia, while HPIV-4 is less common but can still contribute to respiratory disease. Rapid detection guides patient care and infection control.
The kit is validated for RNA extracts from human respiratory tract samples, especially nasopharyngeal swabs.
An internal RNA positive control (IPC), added during extraction and detected in the Cy5 channel, verifies RNA extraction efficiency and identifies possible RT-PCR inhibition.
