Background:  Influenza is an acute infectious disease caused by influenza virus A, B or, to a much lesser extent, influenza virus C. Influenza viruses are enveloped viruses with single-stranded, segmented RNA with negative polarity as genome. These viruses can be found worldwide. Epidemics and pandemics are mainly caused by influenza virus A, due to antigenic drift of the hemagglutinin and neuraminidase molecules. Type B and C influenza viruses are isolated almost exclusively from humans, while influenza A viruses infect a wide variety of warm-blooded animals.

Description:  ViroReal® Assay Influenza A/B is a test, based on one-step reverse transcription real-time PCR, for the detection of the matrix protein gene of influenza A virus and of the hemagglutinin gene of influenza B virus. This test is only intended for analysis of environmental samples in combination with ViroReal® Kit SARS-CoV-2 & SARS (DENV00113), which detects the N gene of SARS-CoV-2, SARS-CoV and SARS-related coronavirus (Sarbecovirus).

PCR platforms: ViroReal® Assay Influenza A/B is compatible with real-time PCR instruments which detect and differentiate fluorescence in FAM, VIC and Cy5 channel (e.g. ABI® 7500 instrument (Thermo Fisher Scientific), QuantStudioTM 5, QuantStudioTM 7 (Thermo Fisher Scientific), Mx3005P® (Agilent), qTOWER3G (Analytik Jena), MIC instrument (bio molecular systems), LightCycler® 480 II (Roche), cobas z 480 Analyzer (Roche)).

The assay detects RNA of influenza A (matrix protein gene) and influenza B (hemagglutinin gene) in environmental samples using one-step real-time RT-PCR.
No, both target genes are detected in the VIC channel, and differentiation between influenza A and B is not possible.
The assay uses an internal RNA positive control (IPC) for extraction and inhibition control, and a positive control for influenza A RNA.
Instruments detecting FAM, VIC, and Cy5 channels such as ABI® 7500, QuantStudio™, MIC, LightCycler® 480 II, and cobas z 480 are compatible.
The LoD95% is 15 copies/reaction for influenza A and 28 copies/reaction for influenza B when tested with synthetic RNA.
The ViroReal® Assay includes only the primer/probe mix and positive control, and is intended for experienced users. In contrast, ViroReal® Kits include additional components such as an internal control and optimized RT-PCR mix.
